Difference of Platelet Rich Plasma Formation by Various Centrifugation Speed and Time in Rab

Abstract
|
|
|
Purpose: The purpose of this study was to examine the platelet concentration and the recovery ratio of PRP by centrifugation time and gravitational force (g) in rabbit.
Materials and Methods: 6 New Zealand white rabbit weighted 2.5~3 kg were divided into three groups. 39 blood samples of 9 ml each were collected. Each experimental groups were divided into three groups by centrifugation time and gravitational force. A group was tested at step 1: 150 g for 20 min, step 2: 450 g for 10 min, B group was tested at step 1: 900 g for 5 min, step 2: 1500 g for 15 min and C group was tested at step 1: 2000 g for 3 min, step 2: 5000 g for 5min. Each groups were compared the platelet concentration and recovery ratio.
Results: After step 1 centrifugation, A group noted 254.2¡¾36.7¡¿103/¥ìl and 65.7¡¾14.3 % in concentration and recovery ratio and came out the lowest. B group noted 578.9¡¾116.9¡¿103/¥ìl and 91.2¡¾13.1% in concentration and recovery ratio and came out the highest. C group noted 247.1¡¾37.4¡¿103/¥ìl and 72.4¡¾6.5% in concentration and recovery ratio. The total platelet counts were 1047, 2316, 1811 respectively and B group was stastically significant(p<0.05). After step 2 centrifugation, platelet counts were 1014.9¡¾168.5¡¿103/¥ìl, 1486.0¡¾266.4¡¿103/¥ìl and 1067.5¡¾259.6¡¿103/¥ìl and recovery ratios were 62.1¡¾9.3%, 65.2¡¾10.7% and 59.0¡¾11.9%. B group shows highest value but no stastical significance(p>0.05).
Conclusion: During the formation of platelet rich plasma in rabbit, the step 1 centrifugation with 900 g for 5 minutes and it shows the highest platelet concentration. After the step 2 centrifugation, although each group shows no significant difference, the recovery ratio came out the highest at centrifugation with 1500 g for 15 minutes.
